Jacob van de Kar was a son of Simon van de Kar and Esther de Groot. He married in Amsterdam on 26 January 1933 to Celine Rubens, daughter of Ruben Rubens and Abigael Rodrigues de Miranda. The couple had two daughters, of whom Elisabeth has survived the Holocaust by going into hiding in Deventer.

Jacob van der Kar studied medicine in Amsterdam, where he sat for his medical finals on 23 November 1929. Jacob van der Kar lived and practised medicine at 77 Pretoriusstraat in Amsterdam.
Jacob van de Kar was a very good chess player. He was a member of the Vereenigd Amsterdamsch Schaak Genootschap. During the games of 1926-1927 he became second after dr. Euwe. Besides chess Jacob van de Kar was also good at playing bridge.
