Ephraim van der Sluis was a son of Joseph van der Sluis and Esther Spaanbroek. His siblings were Simon and Elisabeth, (who have survived the Holocaust), Klara, Gerrit, Jaantje, Sientje, Isak, Leentje, Benjamin en Marcus.
Ephraim married Cornelia Bonewit in Amsterdam on 17 July 1929, a daughter of Levie Bonewit and Sophia Roe. They had a son Levie, who was born in 1917 but who only was legalized at the time of their weddding.
