Salomon Lootsteen was a son of Isaac Lootsteen and Rebecca Ossendrijver. He was born in a family with seven children, of whom one sister has survived the war. The other siblings, Mozes, Kaatje, Clara, Benedictus and Hartog have been killed during the Holocaust.
Salomon Lootsteen married 26 June 1935 in Amsterdam to Rebekka Aardewerk, a daughter of Levie Aardewerk and Kaatje Roeg.
