Jacob Pinkhof first studied pharmacology and later medicine in Amsterdam. He obtained his doctoral degree on 27 June 1919 in Amsterdam at the Faculty of Maths and Physics on a thesis 'On the application of electrometric titrations'. On 9 April 1924, he sat for his medical finals. Jacob Pinkhof specialised in ophthalmics. He settled as a specialist at 20 Jan van Eyckstraat in Amsterdam and he also practised at 18 Pretoriusstraat and at 5 Daniël Willinkplein.
He is the author of the 'Berieth Kodesj. Manual to the regulations on circumcision'.
Jacob Pinkhof was married to Branca Asscher. The couple had three children: Herman (also known as Menachem), Rebekka Roza (also kown as Rivka Soshana) and Adele Sophie (also known as Ada). In September 1942, the family took their own lives. The family is buried at the Jewish cemetery of Muiderberg.
